Myrtleford's first post office operated from the property, adding to its historical significance.
Current owner Leshya Perkins has lived at the homestead for nine years.
"Myrtleford Homestead is located in the heart and hearts of the people of Myrtleford," she said.
Over the years, the homestead has played host to an array of commercial interests including the general store, a tea house and a chiropratic and physiotherapy centre.
It currently houses Myrtleford's healing and massage centre.
The property is zoned mixed use and could be suited to a bed and breakfast operation, Leshya said.
"Myrtleford is so starved of accommodation and this place just screams bed and breakfast," she said.
"The local people are very supportive and very much behind another business going into the property."
Leshya said she was "honoured and privileged" to be part of the history of a very grand old home.
"The next owners will be very lucky to be a part of such a special place in Myrtleford's history," she said.
Myrtleford Homestead boasts four main rooms, a two-roomed maid's quarters, pine floors and ceilings, french doors and four open fireplaces.
The homestead has a veranda on three sides and the property has rear and side access as well as car parking at the rear.
